You can hop on the UP express, be downtown in 20 minutes, and 20 minutes back, 9$ each way.
I would just add that you can also get off at the Bloor GO station and check out the neighbourhoods. It depends on what you're into.
Museums, sports, theatre
Take the up and go downtown. You’ll have plenty of time to explore the harbour front, maybe visit the cn tower, or just hang around the city hall, dundas square, etc. Lots of international culinary going on as well
Ya definitely take the UP express into the city. Very easy way to get to Union Station (in the heart of the financial district). From there you can easily walk along Lake Ontario and the water front (this is where the CN Tower is). Or you can head north a few blocks (walk or subway) to Young and Dundas square - the heart of the entertainment district. You will find lots of restaurants and theaters as well as the famous Eaton Center shopping mall.
Eat a type of cuisine that you've never had before! Easily one of Toronto's strengths is food from every corner of the world in every way from "grandma back home" to highly Westernized versions and creative fusions (though not everything is represented well in the downtown core, i.e. you'd have to travel farther for certain things depending on your tastes which might not work with your schedule).
If you want to try a tourist stop like Yonge + Dundas (imho not exciting: malls are malls with many of the same global brands you already know), though a benefit of that stop is that its pretty central. Usually tourist areas (e.g. CN Tower) are more isolated from real food (just chains and overpriced crap), but there are a few places closeby Yonge/Dundas on the stretch of Dundas -> University as you walk west of Yonge that are OK (more fast and quick vs. great, but lots of cuisines represented). At University+Dundas, a well known fusion called Momofuku is just south on University if you want to try somewhere fancier + pricier. Continuing West along Dundas, Chinatown is just a tad further west, starting around Beverley St. The Art Gallery of Ontario (AGO) is right there between Dundas+McCaul and Dundas+Beverley as you get close to Chinatown (you can check online to see if any current exhibitions are of interest). Dundas and Spadina is the heart of Chinatown. If you are curious to see the University of Toronto, which has a sprawling campus and many historical buildings, it is immediately north of Chinatown, starting at College St and going north up to Bloor St.
As an alternate jump-off from Yonge+Dundas in a different direction, Yonge is also close to Church St which is home to Toronto's well-known gay village (go East from Yonge to Church, then go north from Dundas at Church; anything north of Carleton/College St (it changes names at Yonge) is the village and it will very quickly become apparent as you walk up).
I wouldn't be afraid to pick a direction. By staying on these major East/West (College, Dundas, etc) streets and North/South (Yonge, Church) streets it'd be easy to keep your bearings and close to transit that can get you back to either Yonge/Dundas or to Union Station for the UP express back to the airport. Remember to give lots of extra time for TTC (Toronto transit) which is infamous for delays and holdups especially along streetcar routes.
The U.P train to downtown will get you to my fave tourist experience -- the Ripley's Aquarium. If you have such a place in your City then they must all be similar. But I love the glass tunnels that let you see sharks and other sea creatures swimming around and above you. If you are opposed to captive creatures then skip it. The CN Tower is worthwhile is you want to spend the money and wait in the lineup to reach the highest of heights the City can offer. More cash gets you a walkabout on the outside edge. Worth every penny.
take the UPX train from the airport to downtown.. walk west along King Street to Spadina.. walk north on Spadina in chinatown past Dundas West and take a left turn on St. Andrew and go into Kensington Market.. lots of great coffee places, shops, cafes, etc.
Take the UP express from Terminal 1, which will take you to union station in the heart of Downtown, then from their you can visit the CN Tower or go to Kensington Market.

Next visit queen st w for young crowd and relaxed beautiful stores and international restaurants including Chinatown
Walk 10 minutes to largest shopping centre called EATONS Centre Spend 1 hr - over 500?stores This is the centre of the city called DUNDAS Square
Nowvto go back to Airport, Walk under ground from Eaton Centre to train Station- 15 min From there train to Airport and thank you for visit to Toronto
