That seriously would depend on, what you'd like to see film wise and maybe your budget. There are plenty of studio tours and all; but also, you can easily walk or drive around to see where some films were shot. What aspects of the film industry are you most interested in?
The Burbank Studio tour is about the best for seeing a studio, and having a chance to see a show being filmed. It is near Universal Studio tour, as well as Hollywood. In Hollywood, the Egyptian Theater has some history behind it as the 1st theater in Hollywood. They just upgraded their projection booth so they can show nitrate films. Down the street is the El Capitan. Much more modern, but has the Disney snazz put in it. Of course, the Chinese Theater is there as well, but just the facade. ENJOY ~ Mark, the Adventurer

Check out all the restored old movie theaters along Broadway in Downtown LA, there's always some type of event going on these days. Some offer tours, old movie screenings, and other events.
Hiking up to the Hollywood sign is always fun
For Downtown LA check out The Edison (nightlife) and Skyslide at the U.S. Bank Building (not as thrilling as it sounds, but cheap, and awesome views). The rooftop at The W Hotel (Wilshire) and The Standard (LA or Hollywood) are also pretty amazing. In Hollywood, check out The Wiskey-a-GoGo for it's historic value (The Doors were discovered there) and La Brea Tar-Pits (where we are still finding woolly mammoth and sabre tooth cats under city streets - don't pay, just walk around). For meeting people, head to Venice Beach for all the weirdos, Hermosa or Manhattan for the elite plastic babes and hunks, and Long Beach for the down to earth feels like home type (4th st Arts District for fun casual divey, 2nd St for more college town beach vibe, or Pine Ave for the true urban city life). The Queen Mary is also in Long Beach, and Shoreline Village has some great restaurants and is the beautiful backdrop for CSI Miami and Dexter (both filmed there).
I'm on the west side, and I enjoy all of the following: Cinematique (formerly Silent Movie Theatre), Aero (Santa Monica), Nuart, LACMA (many free and low cost viewings), Landmark (Pico), and there is a nonprofit film house in downtown Santa Monica whose name I don't recall but which is s GREAT cinema, has more variety than anywhere in town, is handpicked, has great (reasonably priced) snacks, and is CHEAPER than any other movie house on the west side. Look it up and it pops right up...I just can't recall the name of it. In fact, I'm gonna go to a movie there tonight now that I thought of it :). It's also close to lower-cost or free street parking, than other things in Santa Monica are.

Studio Audience: If your a Conan fan or a fan of some of the talk shows/game shows, several shoot here and you can get free tickets to be in the studio audience. You get to see how it all works and you'll see some of the other studio show's working around you as you walk in as well. Note that for some of them you need to get them online a week or more in advance. I live in LA and still go see Conan from time to time, here's his website: http://teamcoco.com/tickets
Studio Tours: I'll echo the above statements about studio tours, that's probably the most interesting/informative way of getting an inside look at how a film is made & a history of how film was made in the past.
James Dean/Space: If you're a James Dean fan and/or really into space/science, its a tourist trap, but The Griffith Observatory is amazing and worth it, they just finished new construction (=new, cool exhibits) on it about 2 years ago. The last portion of "Rebel Without a Cause" was filmed here and you can go check out where they shot it. (There's also a bust of him there)
