there are more ways and how you can get here depends on your budget, your time limitations and if you have interest in visiting places in between.
Fastest way would be by PLANE. Virgin airlines is the cheapest flying to and from Alice Springs airport. Prices seem pretty stable and you can find for 300-350 $ https://mobile.virginaustralia.com/virginaustralia/index.html
I wouldn't advise BUS, cause takes long and is not cheap. By the way I think greyhound is the most popular company
By CAR is maybe the most interesting way, but also takes long and makes sense if you find travel mates to share gas money with (and very probably unforgettable moments and experiences). Not very heap though. You can post an ad here on couchsurfing or go through coseats.com.au You'll probably not find a direct lift from Sydney to Alice Springs, but you can split the trip and/or combine different transportation methods.
Another interesting way is by CAMPERVAN RELOCATION (there's a section also on coseats.com.au) Basically you pay some dollars per day, have to give a big deposit (~1000) but they often provide some gas money. Negative side is they require you to relocate the CAMPERVAN in a short time (gets a bit tricky if you want to stop visiting places) I think other companies offering camperrelocations are Apollo, britz and maui (or just type campervan relocations on Google)
Finally the cheapest and most adventurous way would be HITCHHIKING. Takes a bit more time, but unforgettable experiences are granted! http://hitchwiki.org/en/Main_Page
The best solution is almost always a compromise if you don't have particular needs. You can fly to Adelaide in order to cover big part of the distance in short time and then hitchhike, rent a car or camperrelocate from there and stopping every 500-800 km. Interesting to see on the way are the Flinders Ranges near Port Augusta (Quorn, Wilpena Pound, ...) Cooberpedy and surroundings (the "breakaways" Amazing views! "Mad Max" and "Priscilla Queen of the desert" were filmed here) If you stop there I can advise the "oasis tourist park". http://www.cooberpedy.net/attractions Finally before reaching alice you should go to Uluru, Kata Tjuta and Kings Canyon.
