What places should I visit for sure , like a must for one day trip!
I will be in Philadelphia just one day :) so any advice will be highly appreciate it!

Depends on what you are interested in. I wouldn't pass up a trip to the Philadelphia Art Museum, Climb the steps and look at the view of the city. Then head over to get a cheesesteak at Gino's or Pat's. Many people will tell you the cheesesteaks aren't great, but if you are going to Philly you have to go there to at least say you were there.
If you have time it won't hurt to see the Liberty bell
If you stay here for the night, go to the museum steps at night. A great view of the city at night from the top of the stairs. West Philly has a shwerma place called Saad's that has tasty food at a good price.
Pura vida, Steven! Eastern State Penitentiary is really unique to Philly! And the Fairmount neighborhood surrounding it is really charming; plenty of places right in front of the museum for lunch. You could walk there from the Art Museum that others have suggested. But if you would rather do a very traditional Philly tour, I would visit Old City, where the Liberty Bell is and all other historical sites. It'll be closer to the cheese steaks (Pat or Gino) that someone mentioned, although I am definitely one of the folks that thinks their sandwiches are awful. If you're on that side of town and want a cheese steak, go to Jim's on South St. Still very famous but much better quality, and a closer walk from Old City!

If you are an art lover I would not miss the Barnes. You need to schedule your visit there and it's totally worth it. The best impressionist art collection in North America. I would visit the Reading terminal for food. It is a food market with a lot of variety. You should get an Indigo bike (bike share) and go around to old city and the historic sites, I would skip some of the tours as this will take a while. Also visit Rittenhouse square, a nice park to people watch. There are good restaurants near the park (Not sure what kind of food you like). I will tag some good places below for reference. Let me know if you need more details.
100% de acuerdo con Dorothy - Eastern State es una experiencia única de Filadelfia, y Fairmount es un barrio encantador. También está cerca del sendero de río Schuylkill (una parte del parque Fairmount), donde tendrías una vista especial de la ciudad. Después de visitar la prisión, puedes pasar por el sendero al sur a Rittenhouse, donde está la plaza Rittenhouse (uno de los parques más famosos del país). Todo esto está más o menos cerca del estación de tren central, 30th Street Station.
